AB Science announces that CHMP adopted a negative opinion for masitinib marketing authorization in indolent systemic mastocytosis after reassessment

AB Science SA (NYSE Euronext - FR0010557264 - AB), a pharmaceutical company specialized in research, development and marketing of protein kinase inhibitors (PKIs), announces that the Committee for Medicinal Products for Human Use (CHMP) of the European Medicine Agency (EMA) has adopted a negative opinion for masitinib in the treatment of adult patients with smouldering or indolent systemic severe mastocytosis unresponsive to optimal symptomatic treatments. This decision follows the reassessment requested by AB Science after the previous negative opinion adopted by the CHMP in May 2017.

The CHMP considered that there were too many uncertainties that precluded the benefit-risk assessment. These uncertainties related to i) the Good Clinical Practice (GCP) findings that were corrected too late by the Applicant, ii) the conduct of the study that excluded from the claim and the analysis the patients with either cutaneous mastocytosis or non-severe indolent systemic mastocytosis, and iii) uncertainties regarding the long-term safety profile of masitinib for a chronic condition.

Overall, the CHMP considered that the product could be considered as active in the targeted indication and that the study has shown a benefit but did not provide enough robust evidence in the context of a registration based on single pivotal study.

AB Science intends to initiate a confirmatory study in patients with smouldering or indolent systemic severe mastocytosis unresponsive to optimal symptomatic treatment in order to confirm the results from this first pivotal study.

About masitinib
Masitinib is a new orally administered tyrosine kinase inhibitor that targets mast cells and macrophages, important cells for immunity, through inhibiting a limited number of kinases. Based on its unique mechanism of action, masitinib can be developed in a large number of conditions in oncology, in inflammatory diseases, and in certain diseases of the central nervous system. In oncology due to its immunotherapy effect, masitinib can have an effect on survival, alone or in combination with chemotherapy. Through its activity on mast cells and microglia and consequently the inhibition of the activation of the inflammatory process, masitinib can have an effect on the symptoms associated with some inflammatory and central nervous system diseases and the degeneration of these diseases.
